It depends, usually after you've built a little rapport. But if it's someone you already know then right away. You start to 'flirt' when you want to push boundaries a little from casual to sexy.... if she flirts back you've been given the green light to escalate.
Just about anything, a little teasing and innuendo works fairly well, eye contact is a must. If she touches you, then touch her back, go from there, but some chicks just like to flirt, and nothing is going to happen, these gals are great to practice on if you are learning how to do this.
