Yes, but on the flip side... if something goes bad between you and his sister, its:
1) going to be awkward whenever you come over
2) going to make things different between you and him
3) going to make him feel like you go to his house for action instead of to see him.
Then again, if him and his friend were friends, he should understand that he's overprotective and his sister is off limits.
Make a choice man. Out of experience, it'd be way better off if you never knew him and just met her, but you do know him and a lot of bad things can come out of this.
If he's just a casual friend and you don't really care about him, make the move.
But if he's a really good friend, he might get a little upset that his friend is moving in on his defenseless sister, especially if you talk to him about your moves on the ladies. What might even be worse is if you make the move and she rejects you. Then you lose both ways. You can always use "Bros over hos," but that's a little outplayed. You seem committed, so if you want, you should just go for it and risk losing a friend.
