From my experience:

If you've already talking and know each other, then they ask questions related to whether you have a girlfriend / they mention to you they don't have a boyfriend/are single.

If you don't talk to each other and are strangers, you can tell when you look at each other. If she smiles when you make direct eye contact, starts fixing herself etc

A few indicators that she’s available:

Wears sunglasses on top of head
Wears red dress
Red shoes
Red lipstick
Wears heart shaped jewelry or clothing with hearts
